Screening of Suitable Plant Regeneration Protocols for Several Capsicum spp. through Direct Organogenesis

چکیده
Peppers (Capsicum spp.) are recalcitrant to in vitro culture regeneration, making the application of vitro-based breeding strategies difficult. We evaluated impact different combinations auxins, cytokinins and micronutrients on induction direct organogenesis cotyledon hypocotyl explants C. annuum, baccatum chinense. found variation regeneration response among species type explant. In this way, average numbers shoots per explant were, respectively, 1.44 0.28 for 4.17 3.20 0.08 0.00 Out six media, best overall results were obtained with medium Pep1, which contained 5 mg/L BAP (6-benzylaminopurine), 0.5 IAA (indole-3-acetic acid) 0.47 CuSO4, followed by a subculture same supplemented 10 AgNO3 (medium Pep1.2). The result Pep1 + Pep1.2 was using explants, 8.87 grown ones greener tissue, while than explants. Our indicate that there is wide Capsicum terms regeneration. suggest synergistic effect copper silver resulted higher rate Explants transferred media elongation, rooting acclimatization. Although acclimatized plantlets chinense, an improvement these latter stages would be desirable high throughput pipeline. This work contributes protocols specific medium, genotype, reaching levels efficiency required genetic transformation gene editing technologies other crops.

متن کاملDirect Organogenesis and Plant Regeneration from Cotyledons of a Multipurpose Tree, Acacia mangium Willd
Using cotyledons of 2-week old in vitro germinated seedlings, a rapid and efficient regeneration protocol was established for A. mangium on Murashige and Skoog (MS) basal medium. Cotyledons showed better direct shoot organogenesis producing maximum 10.88 ± 0.35 shoots per explant when cultured on MS medium supplemented with 2.0 μM BA and 1.0 μM NAA. Full strength of MS medium supplemented with ...

With the aim of developing an efficient plant regeneration protocol, leaflet explants of three accessions of Arachis villosa Benth. (S2866, S2867 and L97) were cultured on basic Murashige and Skoog medium supplemented with different combinations of plant growth regulators: alpha-naphthalenacetic acid, indole-3-butyric acid, 6-benzylaminopurine, kinetin and thidiazuron. The accession L97 was the...

Koelreuteria elegans, popularly known as “Flame Gold” is an ornamental tree. In vitro callus induction and regeneration from various explants (eaf segments and cotyledonary leaf) were studied on modified MS medium. The highest callus induction rate (80%) and multiplication was obtained in 2 mg/l 2,4-D from leaf segments. Calli transferred in 1.5 mg/l BAP resulted in efficient shoot regeneration...
